What Is PT-141 (Bremelanotide) and How Does It Work?

PT-141 (bremelanotide, brand name Vyleesi) is a synthetic melanocortin receptor agonist that treats hypoactive sexual desire disorder (HSDD) in premenopausal women — the only FDA-approved on-demand peptide for sexual dysfunction. Unlike PDE5 inhibitors that act on blood flow, PT-141 works centrally through MC3R/MC4R receptors in the hypothalamus, activating neural pathways for sexual arousal and desire (Kingsberg et al., 2019, PMID: 31218805).

PT-141 Dosing Protocol for Men and Women
